The mechanism of virus inactivation of plasma:
The methylene blue (a kind of intravenous injection drug enrolled in codex) in plasma has more intense affinity with G-C base pair of nucleic acid and it can be activated by light to an excited state which in turn activate oxygen to yield oxidizing radicals. Such radicals can destroy the nucleic acid, especially the Guanine, so that the virus gene is destroyed and the virus is inactivated. It is confirmed by a great deal of experiments that the methylene blue photochemical method can be used to inactivate HCV and HIV in plasma efficiently (virus titer decreases large than 6??CID50, the recovery rate of main blood coagulate factor is larger than 80 percent, the recovery rate of albumin is large than 90 percent, and the immunocompetence of plasma protein has no change). Validated by National Institute for the Control of Pharmaceutical and Biological Products and the Academy of Military Medical Sciences, our technics of virus inactivation can efficiently inactivate VSV, Sindbis and HIV in plasma and accord with the regulation and standard of the Department of Public Health.
The function of the filter for adsorption of the virus-inactivation additive:
The photosensitive additive (0.9~1.3? mol/L methylene blue) which has the function of virus inactivation is added to the plasma. The plasma containing methylene blue in the illumination bag is illuminated, and the photosensitive additive can inactivate the virus in the plasma. The filter can absorb the residual photosensitive additive and eliminate the leucocytes. The virus-inactivation plasma prepared by our product contains 15 percent residual methylene blue which is the 1/4000 of the minimal quantity allowed for clinical using. The color and appearance of the plasma after filtration is similar with the normal plasma, and it will not bring any adverse psychological burden to patients.
