MeltPro® Mycobacterium tuberculosis Rifampin-resistance Mutation Test Kit
Mycobacterium tuberculosis Rifampin-resistance Mutation Test is based on the Probe-based MeltingCurve Analysis. According to the change in T m value between targets and the corresponding wild-typeprobe sequences, the test detects the mutations in rpoB gene (codons 507~533), which reports rifam-pin-resistance information.
Detection region: mutations in rpoB codons 507~533 (81 bp) (coverage rate: 95% of all rifampicin-resistant mutants)
Principle of Probe-Based Melting Curve Analysis
Probe-based melting curve analysis is based on melting temperature generated by thermal denaturation ofthe probe-target hybrid. Melting curve plot of fluorescence (F) versus temperature (T) is transformed intomelting peak by plotting -dF/dT versus temperature. The hybridization probe with fully matched wild-typetarget gives the highest T m value. With matched mutant-type target, it gives the lower T m value.Mutation is detected as T m deviation (△T m ) compared to the wild-type hybrid.
