Mexedrone is a stimulant drug of the cathinone class that has been sold online as a designer drug. It is the alpha-methoxy derivative of Mephedrone.
Mexedrone acts as a weak serotonin–norepinephrine–dopamine reuptake inhibitor (SNDRI) with IC50 values of 5289 nM, 8869 nM and 6844 nM, respectively, as well as a weak serotonin releasing agent (SRA) with an EC50 value of 2525 nM.

Mexedrone is a compound belonging to the substituted cathinone family of stimulants which first appeared online in 2015.
It has the formal and systemactic IUPAC name 3-methoxy-2-(methylamino)-1-(p-tolyl)propan-1-one. It has a relative molar mass of 207.27 grams and an empirical formula C12H17NO2.
Cathinones are similar in structure to amphetamines. They contain a phenethylamine backbone with a benzene ring substituted with an ethyl chain connected to an amino group with a alkyl substituent at the alpha carbon adjacent to the nitrogen. Cathinones additionally have a ketone substituent at the beta carbon which differentiates them from amphetamines.
Mexedrone specifically contains a methyl substituent at the nitrogen analogous to the amphetamines MDMAand methamphetamine, and a methyl substitution at the para position on the aryl ring similar to mephedrone. Then a methoxy (CH3O) group is bound to the methyl at the alpha carbon. It is this methoxy group that givesmexedrone its name.
Therefore, structurally mexedrone is an alpha-methoxy derivative of the widely known substituted cathinone 4MMC also known as mephedrone. It has been characterised as a weak and non-selective reuptake inhibitor at the dopamine transporter (DAT), noradrenaline transporter (NET) and serotonin transporter (SERT) with IC50 values in the low micro molar range. Additionally it displays weak serotonin releasing character at SERT with an EC50 of 2.5 μM. This activity is similar to the activity of mephedrone which also acts as a serotonin releasing agent.
Increased dopamine and serotonin in the synapse are responsible for the stimulatory, euphoric and entactogenic properties of these compounds.
The long term toxicity and health effects of recreational consumption of mexedrone are unknown. It has a very short period of human use.
Mexedrone is not for human or animal consumption.
A range of physical and cognitive effects have been reported by unverified sources online.
Physical effects of mexedrone are reported as euphoria, enhancement of touch and a warm soft tingling sensation which rises up to a maximum during the peak of the experience. A sedating component distinguishes mexedrone from its close relative mephedrone which is a stimulant. This will discourage physical activity such as dancing.
Negative physical effects which have been reported include dehydration, blurry vision, perspiration, heating of the body and grinding of the teeth in a manner similar to but less severe than MDMA.
Subjective cognitive effects are reported as including a mild euphoria and feelings of happiness and acceleration of thought. There are some reports of auditory enhancement.
Negative after effects are also reported including loss of motivation, difficulty thinking, anxiety, difficulty thinking, depression and irritability.
It is reported as being of a lower potency than mephedrone and producing less stimulation and euphoria.
Stimulants in general may be psychologically addictive and encourage compulsive use. The cathinones have a high potential for abuse. Tolerance develops with repeated use meaning people will use ever increasing doses to achieve the "high". Higher doses put a greater strain on the body and may be toxic.
It is potentially dangerous to mix mexedrone with alcohol, cocaine, tramadol and NBOMes.
