Acrosome reaction occurs during period when the sperms are combined with the pellucid zone of the ovum and before sperms fertilize oocyte by penetrating the membrane of oocyte. The SpermFuncTM ARIC can evaluate the ability of sperm's acrosome reaction by the staining with fluorescence after sperm capacitation and acrosome reaction induced by calcium ionophore A23187 in vitro. The test result can provide the evidence for analysis of infertile pathogeny and selection of the treatment in ART.
When the calcium ionophore A23187 induces the Ca2+ into the inside of sperm, the acrosome reaction happened by the allosterism of sperm acrosome. Since there are glycoproteins in outer membrane of sperm acrosome which can be combined with the lectins specifically, the transformations of the zone in the sperm acrosome can be observed by staining with the lectin fluorescence so that we can evaluate the status of acrosome reaction in sperm.
